Introduction: Periodontitis is a chronic inflammatory disease characterised by inflammation of the tooth supporting tissue. On the other hand, oral cancer is being the major health concern of people worldwide. Head and neck cancers appear to be the 6th most commonly occurring ones. Gene – gene interaction interactome studies pave a great pathway in correlation studies, which could lead to the identification of the hub genes. These hub genes become potential biomarkers and druggable targets that help in the elimination and eradication of the disease. Aim: The current study aims at studying the gene – gene interaction interactome of periodontitis and oral cancer, and at identifying the hub genes of significant importance. Materials and Methods: The GEO2R online analysis tool and NCBI GEO datasets were used to assess the expressed genes. The linking genes were found and classified using GeneMANIA, and the gene network was created by combining genes expressed in Parkinson’s disease and periodontitis. PantherDB pathway was used to conduct a gene enrichment analysis for Gene Ontology (GO) keywords, in order to define the biological significance of the major gene connected to this interactome. Results and discusssion: The differentially expressed genes were imported into the GSEA programme. The gene set database was used to find the hub genes. Out of the 20 hub genes discovered, 10 showed considerable expression. The hub genes expressed underwent gene enrichment analysis, and their biological and molecular processes were investigated. Conclusions: From the results of our study, it can be concluded that a positive gene – gene interaction interactome is observed between periodontitis and oral cancer mediated by integrin signaling pathway. The interlink between periodontitis and Oral cancer via the hub genes, and the signaling pathways could be used for drug targeting, while also acting as a potential biomarker. Further case-control and clinical studies are to be conducted to confirm the common pathway activation in the disease.
